Pacquiao, 31, named Mayweather, Floyd Mayweather Sr., and promoters Oscar De La Hoya and Richard Schaefer as defendants.
The suit claims allegations by Mayweather that Pacquiao used performance-enhancing drugs are false and are intended to hurt his reputation. The suit seeks more than $75,000, plus punitive damages, the Los Angeles Times reported.
On his Web site, Pacquiao restated his claim about a difficulty in taking blood and said he does not want to be subjected to random testing within 30 days prior to the fight. He reiterated he has never used anabolic steroids.
"I don't even know what they look like," he said. "My other concern is how close to the actual fight itself can they take the blood. It is my opinion taking blood from a person can weaken you. I do not want to be in a weakened state when I enter the ring against any fighter."
Pacquiao's record is 50-3-2 with 38 knockouts. Mayweather, who retired in 2007, is 40-0-0 with 25 knockouts.
Plans for a Pacquiao-Mayweather fight in March have been put on hold while the two camps have publicly aired their differences on the matter of testing.
