Cassel tossed for 152 yards and a touchdown and Randy Moss had six receptions for 116 yards and a score for the Pats.
The veteran Brady injured his left knee and left the game in the first quarter.
Sammy Morris rushed for 53 yards and a touchdown for the Patriots (1-0).
Damon Huard threw for 118 yards and a touchdown for Kansas City (0-1).
Chiefs' receiver Dwayne Bowe caught five passes for 49 yards.
