Kevin Kolb, who was the team's starting quarterback when the season opened, and was injured in the first game, will replace Vick. He filled in after Vick suffered a midsection injury last Sunday during the Eagles' 17-12 loss to Washington.
Doctors later said Vick suffered cartilage damage and a small fracture in his rib area.
Vick had taken over for Kolb when Kolb suffered a concussion in the opener. Last week, after the Vick injury, Kolb completed 22-of-35 passes for 201 yards and one touchdown. He also threw an interception.
The Eagles also said cornerback Asante Samuel and wide receiver Riley Cooper, who both suffered concussions in the Washington game, are out for the 49ers contest this week.
